Nature of the research In this study we would like to optimise and validate a digital image analysis algorithm for Ki67 proliferatie-index in NETs based on a virtual double staining for Ki67 and synaptophysin.

Background / introduction
The Ki67 proliferation index is used to grade neuroendocrine tumours (NETs). Manual assessment is time-consuming and prone to intra- and interobserver variability. Especially in the recently categorized group of well-differentiated grade 3 NETs proper assessment of proliferation is crucial, as patients with NETs with a proliferation-index between 20-55% will be treated according to a different adjuvant chemotherapy protocol. Digital image analysis may aid pathologists in assessing the Ki-67 proliferation index in synaptophysin (a diagnostic marker protein expressed by NETs) positive tumour cells. For this study we want to include all the patients from 2014 to 2018 who were diagnosed with a NET. In our research group we recently successfully validated and clinically implemented an algorithm for the assessment of Ki67 in breast carcinoma.1
